Background and objective Uterine leiomyoma is the most common benign tumor in females of reproductive age. However, its causes have never been fully understood. The our study was to analyze causal association between various factors uterine using Mendelian randomization (MR). Methods Genetic variables associated with risk were obtained from genome-wide studies. Summary-level statistical data for FinnGen UK Biobank (UKB) consortium. We used inverse variance weighted, MR-Egger, weighted median methods univariate analysis. Multivariable MR analysis identify independent factors. A fixed-effect model meta-analysis combine results UKB data. Results In data, higher genetically predicted age at natural menopause, systolic blood pressure (SBP), diastolic (DBP), fasting insulin an increased leiomyoma, while menarche a reduced leiomyoma. SBP DBP showed that might be factor SBP, DBP, replicated. result suggested could also affected by polycystic ovary syndrome (PCOS), endometriosis, 2-hour glucose level. Conclusion Our confirmed earlier menstrual age, hypertension, obesity, elevated post-challenge relationship smoking ruled out. addition, later menopause endometriosis found increase PCOS decrease risk.
